Interaction with other medicaments and other forms of interaction In vivo observations Limited clinical data on medicine interactions are available. However, Prograf has been administered together with a great variety of other medicines in clinical trials. Concomitant administration of methylprednisolone has been reported both to increase and decrease plasma levels of Prograf. Imidazole antimycotics such as clotrimazole [from troches], fluconazole and ketoconazole ; , macrolide antibiotics such as clarithromycin and erythromycin, for which up to a 10-fold increase in blood trough levels has been observed ; , danazol and omeprazole have been reported to increase tacrolimus whole blood plasma levels, whereas rifampicin has been shown to decrease tacrolimus whole blood and plasma levels. After combination of either amphotericin B or ibuprofen with Prograf, enhanced nephrotoxicity has been observed. In rats, Prograf decreased the clearance and increased the half-life of pentobarbital and antipyrine. Combination with ciclosporin The half-life of ciclosporin has been shown to increase when Prograf is given simultaneously. In addition, synergistic additive nephrotoxic effects can occur.
